TIR drivers in CGC
Using self-excising cassette (SEC) selection (Dickinson et al., 2015) to generate edits of endogenous genes and introduce transgenes results in an initial knock-in with the SEC (Rol, hygromycin resistant), and a final strain with the SEC excised. We're planning to submit some new strains to the CGC expressing TIR for the auxin-inducible degron system (Zhang et al., 2015). We're trying to determine how the community prefers to use SEC-derived strains from the CGC to maximize utility, but also avoid duplication of strains for the CGC. The initial phenotypic markers in the initial SEC knock-in strains can be useful for crosses, but require SEC excision before experimental use. The SEC-excised alleles can be genotyped by PCR, so skip the SEC excision step before experiments.
