Talk TitleUnderstanding cortical organization using in situ sequencing

Abstract: 
The cerebral cortex is composed of neurons with diverse gene expression and long range projections. Uncovering the organization of diverse neuronal types and their connections provides a foundation for understanding cortical circuits. Towards this goal, my lab develops two complementary approaches to interrogate brain-wide circuit architecture with cellular resolution and unprecedented throughput. First, we develop highly multiplexed barcoded connectomics techniques that can map neuronal projections and synaptic connectivity with cellular resolution and high throughput. Second, we develop high throughput and low cost in situ sequencing techniques that can interrogate both projections and gene expression on a multi-brain scale. As a proof-of-principle, we combine brain-wide in situ sequencing with developmental perturbation to reveal the roles of peripheral inputs in shaping cortical areas at the cell type level. Our work provides a foundation for understanding cortical patterning and variations at unprecedented resolution.
